Pregnenolone: The Forgotten Hormone of Longevity

When most people think about hormones and aging, testosterone, estrogen, and maybe DHEA come to mind. But there’s another hormone that quietly sits at the top of the cascade, influencing nearly everything else: pregnenolone. Sometimes called the “mother hormone,” pregnenolone is the building block from which many other hormones are made — including progesterone, DHEA, estrogen, testosterone, and cortisol.
Like most hormones, pregnenolone levels are high in youth and decline steadily with age. That drop may explain some of the changes we notice in energy, mood, memory, and resilience as the years go by. So, what happens if we support pregnenolone levels as we age? Could this “forgotten hormone” help us stay sharper, calmer, and more vital?
What is Pregnenolone?
Pregnenolone is a steroid hormone made primarily from cholesterol in the adrenal glands, brain, and reproductive organs. From there, it serves as the raw material for many of the body’s most important hormones.
Think of pregnenolone as the first domino in a long chain. When it falls, the rest of the hormones in the cascade are affected. Without enough pregnenolone, the body may struggle to produce adequate amounts of other hormones — and that ripple effect can impact multiple systems.

Available in oral capsules and compounded topical creams. However, oral pregnenolone is subject to first-pass metabolism through the liver, which converts much of it into neurologically inactive compounds. For this reason, I do not recommend oral use. I prefer pregnenolone cream, which bypasses the liver and helps maintain its neurologic activity.
Access
Medical-grade pregnenolone cream is only available by prescription.
Safety
Generally well tolerated, but too much may cause overstimulation, insomnia, or hormonal imbalance.
Medical supervision
Since pregnenolone influences many hormone pathways, it should always be used under the guidance of a clinician experienced in bioidentical hormones.
It’s also important to note that pregnenolone is not FDA-approved for treating specific medical conditions, though it has been studied for several.
Is Pregnenolone Right for You?
